Aladdin's Antigen Retrieval Solution for Floating Sections is designed for antigen retrieval of floating sections, but can also be used for paraffin sections, frozen sections and other samples after fixation with paraformaldehyde, formaldehyde or other aldehyde reagents.This product employs the widely used antigen retrieval reagents such as sodium citrate with a pH of approximately 8.5, which can effectively reduce the risk of fragmentation of floating sections and remove cross-linking between proteins caused by aldehyde reagents, fully exposing epitopes in samples to improve immunostaining results significantly.Normally, paraffin sections require antigen retrieval, whereas frozen sections can be stained without antigen retrieval. This product is particularly suitable for immunostaining of floating sections. Precautions：

The antigen retrieval can be performed in staining jars with staining racks or mailing clips. Plastic staining jars, staining racks and mailing clips can withstand boiling water very well, while glass ones are easily broken due to sudden cold or heat.This product must be diluted 10 times with ddH2O or Milli-Q water prior to use.This product is for R&D only. Not for drug, household, or other uses.For your safety and health, please wear a lab coat and disposable gloves during the operation.

Instructions for Use：

1. Antigen retrieval of paraffin sections.a. Dewaxing: Dewax the section in xylene for 5 min and then replace with fresh xylene for a total of 3 dewaxes in xylene. Then immerse the section in absolute ethanol for 5 min, twice; 90% ethanol for 5 min, twice; 70% ethanol for 5 min, once; and distilled water for 5 min, twice.b. Antigen retrieval: Dilute this product (10X) 10 times with ddH2O or Milli-Q water to obtain Antigen Retrieval Solution (1X). Immerse the sections in preheated Antigen retrieval Solution (1X) and incubate at 80ºC for approximately 30 minutes, then cool to room temperature in approximately 20-30 minutes. Wash 1-2 times with Immunostaining Wash Buffer for 3-5 minutes each time. Subsequent immunostaining steps such as blocking can then be performed.2. For frozen sections.Wash the sections with Immunostaining Wash Buffer for 5 minutes. Immerse the sections in preheated Antigen retrieval Solution (1X) and incubate at 80ºC for approximately 30 minutes, then cool to room temperature in approximately 20-30 minutes. Wash 1-2 times with Immunostaining Wash Buffer for 3-5 minutes each time. Subsequent immunostaining steps such as blocking can then be performed.3. For antigen retrieval in other samples, refer to the above procedures for either paraffin sections or frozen sections.
