Product Description | Aladdin's Deacetylase Inhibitor Cocktail, 100X is designed to maintain the acetylation of histones and other non-histone proteins. It contains 40μM Trichostatin A, 1mM EX-527, 400mM nicotinamide, and 200mM sodium butyrate in 70% DMSO.Cell or tissue extracts contain many endogenous proteases, phosphatases, and deacetylases, which easily leads to degradation or de-modification of protein samples. Therefore, adding appropriate inhibitors is generally necessary for protein extraction. To better detect the acetylation level of intracellular proteins, appropriate deacetylase inhibitors should be added to the cell culture for a period of time before collecting cells for protein extraction. This product can effectively inhibit various deacetylases including class I/II/III HDAC and maintain the acetylation state of proteins.For the protein extraction from different types of samples, aladdin provides a wide variety of protease, phosphatase, deacetylase and other inhibitor cocktails for selection. Please refer to Table 1 in Appendix for details.This product can effectively inhibit various deacetylases, including class I/II/III deacetylases. Among its components, Trichostatin A is a reversible inhibitor of class I/II deacetylase. EX-527 is a selective and reversible inhibitor of SIRT1 and also has a certain inhibitory effect on SIRT2 and SIRT3. Nicotinamide is a reversible inhibitor of Class III deacetylase (Sirtuins, SIRT1-7). Sodium butyrate is a reversible inhibitor of class I/II deacetylase. Please refer to Table 2 in Appendix for specific information of various inhibitors.This product is usually sufficient for preparing 100ml of lysis buffer or cell culture medium.

Instructions for Use: 1. This product is a 100X concentrated stock solution and should be added to the lysis buffer or culture medium at 1:100 before use (e.g., add 10μl of deacetylase inhibitor cocktail to 1ml of lysis buffer). The lysis buffer or culture medium containing the deacetylase inhibitors should be prepared freshly. After preparation, protein extraction from cells or tissues should be performed immediately.
