Summary

Many of the Enterobacteriaceae have partially identical antigenic structures. Because of this one bacterium can cross agglutinate with the corresponding antiserum of another bacterium. An excess of a bacterial antigen is combined with an antiserum that undergoes a cross-aggregation reaction. The common antibody can be removed and the remaining antibody can only react with the specific antigen, this test is called agglutination absorption test.

Principle

Using the agglutination absorption test, monovalent factor sera can be prepared for analysis of bacterial antigenic structures and identification of bacterial types.

Move

1. 1/10 dilution of S. typhi immune serum and S. enteritidis and S. typhi respectively for slide agglutination, can be seen typhoid immune serum and the two kinds of bacteria have agglutination, indicating that the two bacteria have a common antigenic structure, so the occurrence of cross agglutination reaction.

2. add a certain amount of S. enteritidis into 1:10 dilution of S. typhi immune serum, put 37 ℃ water bath in the role of 2 hours, remove the centrifugal precipitation, absorb the supernatant night, discard the precipitate.
3. Agglutinate the absorbed supernatant with diluted S. Enteritidis solution on a slide and observe the results. If there is still agglutination, the supernatant night will be reconstituted with concentrated S. Enteritidis solution and absorbed as above.
4. Repeat the slide agglutination of the absorbed supernatant night with diluted S. enteritidis and S. typhi bacterial fluids, respectively. If there is no agglutination with the former but there is agglutination with the latter, it means that the anti-enteritis antibody in the S. typhi immune serum has been completely absorbed.
