Antithrombin III assay

Summary

The turbidity of the immunocomplex produced by Antithrombin-III (AT-III) and its corresponding specific antibody is measured by transmission method, and its turbidity is directly proportional to the concentration of AT-III in the serum. This experiment is derived from the experimental instruction of the undergraduate 5-year professional laboratory of Mudanjiang Medical College.

Operation method

Antithrombin III Content Measurement Test

Principle

The turbidity of the immunocomplex produced by Antithrombin-III (AT-III) and its corresponding specific antibody was measured by a transmission method, and its turbidity was proportional to the concentration of AT-III in the serum.

Move

1. Manual, semi-automatic determination

Measurement of A-value of each tube with antiserum at 340nm in a water bath at 37℃ for 15 minutes.

Data processing: AT-Ⅲ content (mg / L) - (measured tube A value / reference tube A value) X reference concentration.

Normal value: 210-360 dL

Caveat

1. When the concentration of the specimen is too high, the dilution ratio of the specimen should be adjusted and re-measured.

2. high blood fat. Jaundice and mild hemolysis have little effect on the results of this method.

3. The linear range of this method is 0-560mg/L-.

4. The parameters can be adjusted according to the different models of automatic analyzers.Clinical significance: High AT-III content: seen in hemophilia, aplastic anemia, heart valve disease, uremia, renal transplantation, etc. Decreased AT-III content: seen in DIC, liver disease, toxemia thrombosis, oral contraceptives and other diseases. Oral contraceptives and other diseases.
