Program 19.1 Cleaning of Incubators

Move the culture to another incubator, turn off the power to the empty incubator, then scrub it with detergent and ethanol and turn on the heat switch in order to dry the incubator. The tray was replaced with fresh water and CO2 was reintroduced.
Materials and Instruments
Water with 2% Roccal or similar fungal inhibitor
Decontaminant 70% ethanol
Alternative CO2 warmer or plastic box and tape for closure
Move
1. Move the entire culture to another CO2 incubator, or place the culture in a sealable container (e.g., desiccator), ventilate with CO2 gas, and place in a normal incubator or greenhouse.
2. Disconnect the warmer from the power supply when ready to be cleaned.
3. Remove all partitions and water trays and removable parts from the incubator.
4. Clean the interior of the terrarium with a solution containing a decontaminant (10 % Roccal, or other bacteriostatic decontaminant), scrubbing every nook and cranny as much as possible.
5. Rinse with clean water.
6. Scrub the inside of the tank with 70 % ethanol.
7. With the door open, heat (no CO2 gas) until the chamber is dry.
8. Scrub the partitions and parts with detergent, then rinse with water and scrub with 70 % ethanol.
9. Put the partition and parts back into the incubator.
10. Put back the water tray and fill it with water containing 2 % Roccal.
11. Close the door and turn on the CO2 gas.
12. When the temperature and CO2 have stabilized, return the culture to the incubator.
