Detection of IL-18

Summary

Newly synthesized IL-18 is a precursor molecule to the signal peptide, which is enzymatically cleaved in order to activate it. In this ELISA method, in order to detect only mature (activated) IL-18, the coated antibody must be specific for activated IL-18. Some cells (e.g., human P B M C ) can also secrete IL-18 precursors under normal conditions, and culture supernatants from these cells can be used to detect one or both forms of IL-18. mixtures of anti-human or mouse IL-18 monoclonal antibodies can be used to detect the IL-18 precursor and/or mature form. In this protocol, the antibody to human IL-18 is specific for activated IL-18 only, and the anti-mouse IL-18 antibody detects both forms of mouse IL-18.

3 . 洗 板 4 次 ：快速吸去或倒去孔内容物，再加满洗液。 4 . 每 孔 加 200/xl封闭液， 37°C孵 育 I h 或 4°C放置过夜。 5 . 用检测缓冲液稀释IL-18参考标准品到lOOOpg/m l 。取 5 支 1.5m l 微量离心管， 2 倍 稀释标准品到31. 2pg/ml。 6 . 用检测缓冲液稀释待测样本。样本起始浓度应为1〇〇 〜l〇〇〇pg/m l 。取 5 个 1.5m l 微 量离心管， 2 倍稀释样本。 样 本 来 源 不 同 ，稀 释 度 也 不 同 。研 究 者 可 根 据 经 验 或 做 预 实 验 加 以 确 定 。 7 . 如 步 骤 3 , 洗涤抗体包被的检测板4 次。 8 . 每 孔 加 IOOm I稀释的IL-18标准品或稀释的样本，每个稀释度加2 孔 。至少另有2 孔 各 加 100/^1检测缓冲液以决定背景。室 温 （20〜25°C ) 放 置 2h 。如步骤3 , 洗涤检测 板 4 次 。 9 . 用偶联缓冲液稀释过氧化物酶标记抗体（159-12B 或 93-10C ) 到 0.5Mg/m l 。每孔加 IOOm I，室 温 （20〜25°C ) 放 置 2h 。如 步 骤 3 ，洗涤检测板4 次 。 10 . 最后一次洗板后，立即向每孔加入100^1显色底物，室温放置30min。 1 1 . 每孔加入IOOiUl终止液，室温中， 30m i n 内 在 E L ISA读板仪上读板，根据底物特点 选择适当的滤光片。 1 2 . 根据标准品的光吸收值（O D ) 绘制标准曲线（O D 值 对 IL- 1 8的 pg/m l绘图）。 根据标 准曲线计算待测样本中IL- 1 8的 含 量 （细胞因子ELISA的结果计算见单元5. 2)。
