Product Description

Aladdin's Methyl Green-Pyronin Staining Solution is commonly used to stain cells or tissue sections, based on the methyl green dye that can bind to DNA and stain nucleus in green or blue-greenish, and the pyronin dye that can bind to RNA in cytoplasm and nucleolus and stain them in red.Different from most other MGP Staining Solutions, this product does not contain toxic methanol.This product can be used in combination with immunofluorescence staining or immunohistochemistry staining. The MGP staining can be performed either before fluorescence stains or after the immunohistochemistry staining.One package of this product is sufficient for staining at least 200 samples.


Precautions

When there is a large number of samples, staining racks or tanks produced by can be used for easier operation.For first-time users of this product, we recommend performing a preliminary test with 1-2 samples.This product is for R&D only. Not for drug, household, or other uses.For your safety and health, please wear lab coat and disposable gloves during the operation.


Instructions for Use

1. Sample preparationa. For paraffin sectionsDewax paraffin sections in xylene for 5-10 minutes;Replace with fresh xylene and dewax for another 5-10 minutes;Anhydrous ethanol for 5 minutes;90% ethanol for 2 minutes;70% ethanol for 2 minutes;Distilled water for 2 minutes;Proceed to step 2.b. For frozen sectionsWash frozen sections with distilled water for 2 minutes;Proceed to step 2.c. For cultured cellsFix cells with 4% Paraformaldehyde Fix Solution for more than 10 minutes;Wash cells with distilled water for 2 minutes;Replace with fresh distilled water and wash for another 2 minutes;Proceed to step 2.2. Methyl Green-Pyronin staininga. Stain samples with MGP Staining Solution for 5-10 minutes (The duration of time can be adjusted appropriately).b. Rinse with distilled water 2-3 times for a few seconds each time.3. Dehydration, clearing, mountinga. Dehydrate samples with acetone thrice (3 dips) for 1-2 seconds each.b. Rinse with acetone/xylene (1:1) twice for 1-2 minutes each.c. Transparent samples with xylene for 1-2 minutes.d. Replace with fresh xylene and clear for 1-2 minutes.e. Use neutral gum or other mounting medium to mount the slides.f. Examine samples by light microscopy.
