Product Description

Aladdin's Modified Giemsa Staining Solution, also known as Wright-Giemsa Staining Solution, can be used to stain cells, blood, bone marrow smear, tissue sections, or other samples. According to the chemical properties of the cell components, this product can stain cytoplasm into pink or blue and nucleus into purple or blue-violet, which facilitates the observation of intracellular structures and the identification of abnormal changes in cells.Giemsa stain, named after Gustav Giemsa who is a chemist and bacteriologist in Germany, was originally used for the diagnosis of parasites in malaria patients and later for histopathology and cytogenetics due to its high-quality staining of chromatin and nuclear membranes. It stains cytoplasm well with clear displaying of intracellular structures, but the nucleus is darkly stained with poor demonstration of nuclear structures. This modified Giemsa Staining Method combines the Giemsa staining method with the Wright staining method to achieve a better staining of both cytoplasm and nucleus. Precautions

Blood smear or bone marrow smear should be uniform in thickness to enable better staining and photographing.This staining solution after dilution should have a metallic luster, otherwise it may be invalid.This product should be diluted with ddH2O, deionized water, or ultra-pure water. Do NOT use PBS for the dilution. At low temperatures, precipitation may occur in Giemsa Staining Solutions, which generally does not impact on the staining result.Giemsa Staining Solution is toxic to human body and flammable. Please take effective measures to avoid inhalation or direct contact with the skin.This product is for R&D only. Not for drug, household, or other uses.For your safety and health, please wear a lab coat and disposable gloves during the operation.


Instructions for Use

1.Reagent preparationPreparation of Modified Giemsa Staining Solution (1X): According to the number of samples, prepare an appropriate amount of Modified Giemsa Staining Solution (1X) by diluting the Modified Giemsa Staining Solution (20X) with PBS or ddH2O. For example, add 1ml of Modified Giemsa Staining Solution (20X) to 19ml of PBS and mix well. Note: The Giemsa Staining Solution (1X) should be prepared freshly.2.Sample preparationa.For paraffin sections:Dewax paraffin sections in xylene for 5-10 minutes;Replace with fresh xylene and dewax for another 5-10 minutes;Immerse in absolute ethanol for 5 minutes;Immerse in 90% ethanol for 2 minutes;Immerse in 80% ethanol for 2 minutes;Immerse in 70% ethanol for 2 minute;Proceed to step 3.b.For frozen sections:Wash frozen sections with distilled water for 2 minutes;Proceed to step 3.c.For blood or bone marrow smear:Make blood smears or bone marrow smears according to conventional methods and air-dry;Fix with 70% ethanol for 10 minutes;Proceed to step 3.d.For cultured cells:Fix cells in 70% ethanol for 10 minutes;Proceed to step 3.3.Staininga.Add an appropriate amount of Modified Giemsa Staining Solution (1X) to the section or smear, and stain for 45 minutes. Note: If the staining result is too dark or too light, adjust the concentration of the staining solution or the staining time.b.Wash the slide thoroughly with distilled water from one side, and then examine by microscope after air-dry.Related Products:Cat.
