It not only eliminates mycoplasma that already exists in cells or culture medium, but also prevents mycoplasma contamination.Mycoplasma Prophylactic and EliminationReagent II mainly contains tetracycline and diterpenoid antibiotics that efficiently target mycoplasma. These two antibiotics inhibit protein synthesis through different mechanisms. Tetracycline antibiotics bind to the A sites of ribosomal 30S subunits, inhibiting peptide prolongation by preventing the binding of aminoyl-tRNA to this site. Diterpene antibiotics bind to ribosomal 50S subunit and inhibit peptidyl transferase activity. This product is only effective to mycoplasma and bacteria, and basically has no effect on the biological functions and growth of eukaryotic cells.The reagent stock is provided in solution. Instructions for Use:
1. Reagent preparation:Melt the reagent and dissolve completely. Take appropriate amount for instant use, aliquot the rest and keep at -20℃ for long-term storage.Note: Small amount of precipitation in the reagent after melting has no effect on its activity. Vortex to promote its dissolution.2. Prevention or inhibition of mycoplasma:Add the reagent to cell cultures at a dilution of 1:500. Mix well and culture cells under normal conditions. In general, it can effectively prevent mycoplasma contamination or inhibit mycoplasma proliferation.3. Elimination of mycoplasma:a. If cells are contaminated with mycoplasma, cultivate cells in culture medium supplemented with this reagent at a dilution of 1:100. Refresh the culture medium every 2-3 days. If mycoplasma is eliminated after 2-3 weeks, the culture medium can be replaced with normal culture medium without or with this reagent at 1:500 dilution.
