Product Description | Aladdin's Phosphatase Inhibitor Cocktail C (50X) can be used to inhibit phosphatase activity in protein extracts of animal cells or tissues, fungi, bacteria, and plants. It contains 1.25mM (-)-p-bromotetramisole oxalate, 250μM Cantharidin, and 250nM Microcystin-LR in DMSO, with an inhibitory effect on alkaline and Ser/Thr phosphatases such as PP1 and PP2A.Many endogenous proteases and phosphatases are commonly present in cell or tissue extracts, which easily leads to degradation or dephosphorylation of protein samples. Therefore, adding appropriate inhibitors is generally necessary for protein extraction.Different types of protein extracts contain different types of proteases. For example, there is high abundance of serine proteases in mammalian cell extracts, more serine proteases and metalloproteases inbacterial cell extract, and serine, cysteine and metalloproteases in mammalian tissue extracts. Plant extracts often contain large amounts of serine and cysteine proteases, and some contain aspartic proteases. Therefore, aladdin developed a variety of inhibitor cocktails for the protein extraction from different types of samples. Please refer to Table 1 in Appendix for details.This product can effectively inhibit various phosphatases, such as serine/threonine, tyrosine, acidic and alkaline phosphatase, in protein extracts to maintain the phosphorylation state of proteins. Among its components, (-)-p-bromotetramisole oxalate is an irreversible inhibitor of alkaline phosphatase. Cantharidin is a reversible inhibitor of protein phosphatase 2A with IC50 of 40nM. Microcystin-LR is a reversible inhibitor of protein phosphatase 1 and protein phosphatase 2A, with IC50 of 1.7nmol and 40pmol, respectively. Please refer to Table 2 in Appendix for specific information of various inhibitors.This product is usually sufficient for preparing 100ml of lysis buffer.

Instructions for Use: This product is a 50X concentrated stock solution, and should be added to lysis buffer at a ratio of 1:50 for use (e.g., add 20μl of phosphatase inhibitor cocktail to 1ml of lysis buffer). The lysis buffer containing phosphatase inhibitors should be freshly prepared. After preparation, protein extraction from cells or tissues should be performed immediately.
