Product Description

PI is a nuclear staining reagent that can stain DNA. Like EB, PI intercalates into double-stranded DNA and releases red fluorescence. It is often used for the detection of apoptosis or necrosis, and is often used for flow cytometry analysis. Although PI cannot pass through living cell membranes, it can pass through damaged cell membranes to stain nuclei. PI is often used with Calcein, AM, Hoechst 33258 or Hoechst 33342, etc. It can stain live and dead cells at the same time. The excitation and emission wavelengths of the PI-DNA complex are 535nm and 615nm, respectively.

Dyeing process

(1) Prepare 10-50μM PI solution with PBS or appropriate buffer.

(2) Add 1/10 of the volume of the medium to the cell culture medium (1/10 concentration of PI buffer can also be used to replace the medium).

(3) Incubate the cells at 37°C for 10-20 minutes.

(4) Wash the cells twice with PBS or a suitable buffer.

(5) Observe the cells with a fluorescence microscope with a 535nm excitation wavelength and a 615nm emission wavelength filter.

Storage conditions: Store at -20°C dry and dark, valid for one year.
Precautions

MSDS shows that propidium iodide PI is the same as EB mutagenic to germ cells and is a carcinogen. Please take strict care to protect it!

Propidium iodide PI is a Germ cell mutagenicity germ cell mutant; In vitro tests showed mutagenic effects!
