SDS-PAGE Sample Buffer (Reduced, 5×) is a 5-fold concentrated SDS-PAGE gel electrophoresis buffer with bromophenol blue as dye, which is used for protein sample preparation and sampling by reduced SDS-PAGE. The buffer contains DTT, which can break the intra-chain disulfide bonds and inter-chain disulfide bonds of protein molecules, so that the protein subunits connected through the disulfide bonds are separated from each other, and then clear protein bands are shown on the electrophoresis gel after staining with Caumas Brilliant Blue.
Caveat
1. Thaw at room temperature or 37-40°C for a few minutes before adding samples and then shake gently to ensure that the solution is well mixed.
2. This reagent contains DTT, please wear lab coat and disposable gloves when handling.

Procedure
1. Mix the SDS-PAGE upsampling buffer (reduced, 5×) with the protein samples in a 1:4 ratio.
2. Heat the protein sample in a boiling water bath for 3-5 minutes.
3. Allow the protein sample to cool to room temperature after sufficient denaturation and centrifuge at less than 3000 rpm for 30 seconds.
4. After centrifugation, take an appropriate amount of supernatant and add it directly to the SDS-PAGE gel wells for routine electrophoresis.
