Improved TOMEI (iTOMEI)" is a plant tissue-clearing method that specializes TOMEI for fluorescent protein observation by Prof. Sakamoto et al. iTOMEI enables simply transparency and clearly detecting fluorescent protein. This product is a decoloring reagent for iTOMEI.
Description This product enables transparency by simple osmosis in just a few days (more than 2 days) and keeps the fluorescence of a fluorescent protein such as GFP or tdTomato co-expressed as a reporter gene. The mounting solution is also required to complete the iTOMEI method. Please check directions for use for necessary reagents. Directions for Use Required reagents; Paraformaldehyde (PFA) PBS Stains* (option) Mounting solution** ==Process outline (All operations are performed at Room temperature)=============== Tissue fixation Reagent : 1% PFA in PBS Time : 1hr PBS Wash ×3 Decoloring Reagent : iTOMEI-D (TCI Product No. T3940) Time : 24hr PBS Wash ×3 (Optional process Staining*, PBS Wash ×3) Clearing (RI matching) Reagent : Mounting solution** Time : Operation time is determined by the mounting solution. Mounting Reagent : Mounting solution** * The treatment time of DAPI staining is for 30min with 5 µg/mL and the treatment time of Calcofluor White staining is for 10min with 1 g/L of Calcofluor White M3R and 0.5 g/L of Evans Blue, but their adjustments are needed for the purpose. ** Mounting solution : Please use the suitable mounting solution for objective lens.
