UltraBio™
Basement Membrane Matrix, also known as Extracellular Matrix (ECM) or Basement Membrane Extract (BME) gel, is free of lactate dehydrogenase elevating virus (LDEV). Matrix gel is widely used in angiogenesis, tumor cell migration, in vivo tumor modeling, 3D tumor cell models, and organoid studies. This product has good performance and can be used as a substitute of Corning® Matrigel®. The matrix gel is a basement membrane matrix extracted from the Engelbreth-Holm-Swarm (EHS) mouse tumors rich in extracellular matrix proteins. Besides the main component laminin, it also contains type IV collagen, heparan sulfate glycoprotein, entactin, various growth factors, tissue fibrinogen activator and trace amounts of matrix metalloproteinases.
Aladdin’s UltraBio™
product series are free of LDEV virus or other viruses of mouse and tumor origin. Under room temperature conditions, matrix gels can rapidly polymerize to form biologically active three-dimensional (3D) culture matrix that mimics the structure, composition, physical properties and functions of cellular basement membrane in vivo, thus promoting the attachment, maintenance, growth and differentiation of different cells such as vascular endothelial cells, epithelial cells, hepatocytes, neurons, melanoma cells, thyroid cells and hair follicle cells, by which cell morphology, biochemical functions, migration, invasion and gene expression can be investigated.
Aladdin’s UltraBio™ product series include Standard Formulation, Growth Factor Reduced (GFR), High Concentration (HC), High Concentration & Growth Factor Reduced (HC, GFR), for Stem Cells, for Organoids, and many other types of matrix gels with different growth factors, protein levels, and different applications. The Matrix-Gel™ products without phenol red are suitable for fluorescent and colorimetric assays. The matrix gel with growth factors reduced (GFR) is suitable for experiments requiring highly defined compositions, such as studies of cell signaling pathways and cytokines, while the matrix gel with high protein concentration (HC) is suitable for studies of angiogenesis, tumor cell migration and in vivo tumor models. An appropriate matrix gel can be selected by referring to the table below.

The composition of this product is similar to that of conserved, early developing basement membranes. The main components of this matrix gel are approximately 60% laminin, 30% type IV collagen and 8% entactin [7-8], where entactin is a bridging molecule that interacts with laminin and type IV collagen to contribute to the formation of extracellular matrix. Besides the main components, the matrix gel also contains some proteoglycans and growth factors, such as perlecan, vascular endothelial growth factor (VEGF), transforming growth factor-β (TGF-β), epidermal growth factor (EGF), insulin-like growth factor (IGF), fibroblast growth factor (FGF), tissue fibrinogen activator (PLAT) and growth factors naturally present in tumors, as well as residual matrix metalloproteinase derived from tumor cells [9-10]. This matrix gel rapidly polymerizes at room temperature. The matrix gel will start to polymerize above 10℃. At 22-37℃, the large molecules of the matrix gel can chemically bond to each other, promoting rapid polymerization to form a 3D gel matrix. However, at low temperatures (e.g., 4℃), the matrix gel is in a liquid state because there is not enough energy to prompt chemical bonding. This matrix gel can be used for different experimental purposes. The higher the protein concentration of the matrix gel, the higher the viscosity. The protein concentration of non-high concentration matrix gel and high concentration matrix gel is 8-13mg/ml and 16-26mg/ml, respectively. The minimum protein concentration of matrix gel is 3mg/ml, but to avoid incomplete gel polymerization, the final protein concentration of matrix gel used for in vivo studies should be 4mg/ml at least. This matrix gel is subjected to strict quality control and is of high quality. Each batch of related products is strictly tested for protein concentration, endotoxin, gel, angiogenesis, 3D culture of tumor cells, organoid culture, bioactivity, and sterility (bacteria, fungi, viruses, mycoplasma), etc.
Precautions: To ensure the best performance of this product, aliquot this product at the first thawing to avoid repeated freeze-thaw. The 1ml package is also available from aladdin. This product must be stored at temperatures below -20℃ in the dark. It is not recommended to store this product in a frost-free refrigerator because the automatic defrost function of the frost-free refrigerator may affect the temperature inside the refrigerator. This product should be used aseptically to avoid microbial contamination. After thawing, place this product in a sterile area and spray 70% ethanol on the top to sterilize. Thawed matrix gels can remain liquid for about 30 days when placed in a 4℃ refrigerator or on ice in the dark, but it is still recommended to make aliquot right after thawing. Due to the actions of carbon dioxide, bicarbonate, and phenol red (if present), the color of this product may change during freeze-thaw, which is normal and have no effect on its performance. When handling this product, keep hands away from matrix gel part of the bottle to avoid polymerization due to high body temperature. Hold the bottle/tube above the matrix gel with figures. After the experiment is completed, the remaining matrix gel should not be kept for future use. This product is for R&D only. Not for drug, household, or other uses.
