    Precautions

    QuickBlock™ Blocking Buffer is nonreusable generally. But for some primary antibodies with high signal-to-noise ratio, such as primary antibodies against internal reference proteins, this product can be reused 2-3 times. Do not mix the recovered blocking solutions with unused ones.Usually, it takes 5-15 minutes to block PVDF or NC membranes. For some antibodies with very high backgrounds, prolong the blocking time to 30-60 minutes, or overnight at 4ºC if necessary.Because no blocking reagent is optimal for all experimental systems, a proper blocking buffer should be selected base on the special requirement of your experiment. provides a variety of blocking solutions that you may compare and select.Use flat-tip tweezers to gently handle PVDF and NC membranes on the corners or edges to avoid producing any scrapes, creases or indentations on the surface of membrane.PVDF membrane, once infiltrated and activated, should be kept wet by placing the membrane in Western transfer buffer or washing buffer according to the Western Blot procedures. Otherwise, an abnormal background that is hard to be blocked may be produced.This product is for R&D only.     Instructions for Use

    1. Procedure for blocking membranesa. After completing the protein transfer, wash the membrane with Western washing buffer for 1-2 minutes.b. Based on the size of the membrane, add a sufficient volume of QuickBlock™ Blocking Buffer (TBS) in a petri dish or other appropriate container. For regular Western Blot, use 10ml of Blocking Buffer for a membrane of 6.6×8.5cm. c. Clamp one corner of the membrane with a flat-tip tweezer and place the membrane in QuickBlock™ Blocking Buffer (TBS). Make sure that the membrane is covered completely by the Blocking Buffer. Incubate for 10 minutes with agitation on a horizontal shaker. The incubation time can be from 5 to 15 minutes. Generally, 10 minutes of blocking can achieve better blocking results than 1 hour’s blocking with BSA.d. The blocked membrane can be used for subsequent procedures such as primary antibody incubation. Procedure for blocking in IF, IHC, or other experimentsIn the related experimental procedures, simply replace the traditional blocking solution with QuickBlock™ Blocking Buffer (TBS) and reduce the duration of blocking to 10 minutes. We have tested multiple primary antibodies and found that similar blocking results were obtained when the blocking time ranged from 10 minutes to 20 minutes. The results of blocking for 10 minutes with this product were equal to or significantly better than that obtained from the conventional blocking methods.
