Product Description

Aladdin's UltraBio™ Green Fluorescent DNA Loading Buffer is used for loading DNA samples onto agarose or polyacrylamide gels for gel electrophoresis. It contains NA-Green(Nucleic Acid Green) fluorescent dye that binds to nucleic acids at the moment of mixing and emits Green fluorescence.This product is easy to use. A simple three-step procedure includes adding 1/6 volume of 6X DNA Loading Buffer to DNA, loading onto gel and electrophoresis. There is no need to add any dye in agarose gels or stain gels after electrophoresis.This product is safe, sensitive, and highly stable. NA-Green is a significantly safer alternative to ethidium bromide (EB), with very low mutagenicity and no cytotoxicity. It allows detection of >0.6ng of DNA, and has good thermal and photo-stability. Please see Figure 1 for the sensitivity of this product.Figure 1. The actual results may vary due to differences in DNA quality and detection instrument used.Fluorescence detection system for SYBR Green or EB can be used for this product. Green fluorescence is detected on DNA samples stained by this product using blue light at a wavelength of about 500nm or UV light at 260nm. If using the detection system for EB dye, UltraBio™ Red Fluorescent DNA Loading Buffers containing NA-Red dye are recommended, although this product containing NA-Green can also be used, with a 4-5 times lower detection sensitivity. For Gel Imaging Systems with filters for both EB and SYBR Green, this product is recommended.This product uses bromophenol blue as an indicator for easy visual tracking of DNA migration during electrophoresis. Bromophenol blue comigrates with 200-500 bp DNA fragments, which has more or less interference with the observation of DNA fragments around this size. When it happens, please use Aladdin's UltraBio™ Green Fluorescent DNA Loading Buffer (6X, Red) .UltraBio™™ Green Fluorescent DNA Ladders should be used in parallel with this product. You can also use the regular DNA ladder supplemented with an appropriate amount of Aladdin's UltraBio™ Green Fluorescent DNA Marker dye (50X). Please refer to Figure 2 for the stain result of DNA samples loaded with this product and the UltraBio™ Green Fluorescent DNA Ladder .Figure 2. Precautions

This product should be kept in the dark. Exposure to regular indoor light during the electrophoresis does not cause any change in its performance.This product can be added directly to DNA samples for loading and electrophoresis. Agarose gels after electrophoresis are not recommended for reuse.The gel can be observed directly after electrophoresis using a gel imager. If the background is too high, the gel can be washed with nuclease-free water to reduce the background.This product can stain dsDNA, ssDNA, and RNA.When smear or unsatisfactory band separation is observed, the following methods can be attempted: use freshly prepared electrophoresis buffer; reduce the loading amount of DNA; reduce the agarose concentration; use a longer gel; decrease the voltage for electrophoresis and extend the time of electrophoresis to improve DNA separation, etc.This product is compatible with commonly used electrophoresis buffers, such as TAE and TBE.This product is for R&D only. Instructions for Use

1. Prepare agarose gel with appropriate concentration. Do not add any dye to the gel.2. Mix the 6X DNA Loading Buffer well before use. Add 1/6 volume of 6X DNA Loading Buffer to your DNA sample, mix well and load directly onto agarose gel for electrophoresis. Note 1: InstantView™ Green Fluorescent DNA Ladders ( Green Fluorescent DNA Loading Buffer (6X, Red).3. After electrophoresis, use a Gel Imager System for observation or taking photos.
