Aladdin  UltraBio™ TBE Precast PAGE Gel is a safe, ready-to-use, and high-quality gel with regular size. It is primarily used for high-resolution separation or purification of 10-3000bp double-stranded nucleic acids or 10-3000nt single-stranded nucleic acids, such as digested DNA with restriction enzymes, PCR products, primers, and RNA for Southern analysis. This precast gel has a stacking gel of 1.5cm in height. The nucleic acid bands after electrophoresis are flat, clear, fine, and sharp, with minimal edge effects. Additionally, this product employs the high-quality glass plates to reduce non-specific nucleic acid adsorptions, with better resolution effects than home-made TBE PAGE gels.

Aladdin provides a variety of UltraBio™ TBE Precast PAGE Gel at different concentrations. Gradient gel is available in concentration of 4-20%. Fixed gels include 5%, 10%, 12% and 15% gels. Please refer to the table below for parameters of UltraBio™ TBE Precast PAGE Gel.

This precast gel contains a 4% stacking gel with a height of 1.5cm to ensure sharp nucleic acid bands can be obtained.

The ratio of acrylamide to bisacrylamide in this precast gel is 29:1, and the gel thickness is 1.5mm. The maximum loading volume per well of 10-well and 15-well gels is 60μl and 30μl, respectively. The gel plate size is 98×84×4.1mm (width × height × thickness) with a gel size of 81×74×1.5mm (width × height × thickness).

Polyacrylamide gel electrophoresis (PAGE) is a widely used technique in life science research for the separation, purification, assay, identification, and molecular weight analysis of proteins, nucleic acids, and protein-nucleic acid complexes. The commonly used Western blot technique is based on PAGE.

Nucleic acid electrophoresis is a commonly used approach for resolving DNA or RNA fragments based on length. The nucleic acids loaded into gel wells migrate toward the anode in the presence of an electric field due to their negatively charged sugar-phosphate backbones. Nucleic acid fragments of different sizes can be separated during electrophoresis as they migrate at different rate, with longer fragments migrating more slowly than smaller fragments, thus shorter fragments are closer to the anode than longer ones after electrophoresis. For nucleic acid electrophoresis, two types of gels are commonly used: agarose gels and polyacrylamide gels. Compared to agarose gels, polyacrylamide gels have the advantages of higher resolution and sensitivity, lower background staining, less sample volume required, efficient blotting, easier DNA extraction from gels, no interference with enzyme reactions, and higher accuracy and reproducibility. PAGE gels commonly used for nucleic acid electrophoresis include TBE PAGE gels, TBE-Urea PAGE gels and EMSA PAGE gels. Among them, TBE PAGE gels are prepared from TBE buffer (Tris-boric acid-EDTA), and are mainly used for native electrophoresis of DNA or RNA fragments.

This product is prepared with high-purity Tris, boric acid, EDTA and other reagents. It has strong buffering capacity and is suitable for long time electrophoresis, ensuring high resolution and good separation of nucleic acids. Strict quality control ensures that the gel and buffer are free of DNase and RNase.

Selection of 10-well and 15-well precast gels: Nucleic acid bands obtained from 10-well precast gels are flatter and sharper. However, when the number of samples to be examined is large or quantification of nucleic acids is required, we recommend using the 15-well precast gel which has greater throughput and is more convenient for statistical quantitation analysis

This product is safe and convenient to use. There is no need for cumbersome gel preparation and casting, thus avoiding contact with toxic and irritating reagents. After electrophoresis, the gel can be easily removed from the glass plates.

The quality of this product is high and stable, with high consistency between batches. It uses high-quality glass plates to reduce adsorption of nucleic acids significantly, resulting in better electrophoresis performance. This product provides excellent electrophoresis results. It offers excellent nucleic acid resolution. Smooth, clear, fine, and sharp nucleic bands can be obtained.

The electrophoresis time is short. The native electrophoresis can be completed in 50-75 minutes at 150V. The run time can vary depending on experimental requirements.

The gel is easy to remove from the glass plates. Users only need to light scrape one side of the glass plate with a blade, and the glass plates can be easily opened without the need for special lifting tools.


Precautions:

This product should not be stored below 0℃ to avoid freezing and cracking.

The electrophoresis buffer in the inner chamber of the electrophoresis tank and the transfer buffer should be freshly prepared. Low-quality buffers, recycled buffers, or buffers that have been kept for too long period of time will result in poor protein resolution and transfer.
