Protein Function
Responsible for the deacetylation of lysine residues on the N-terminal part of the core histones (H2A, H2B, H3 and H4). Histone deacetylation gives a tag for epigenetic repression and plays an important role in transcriptional regulation, cell cycle progression and developmental events. Histone deacetylases act via the formation of large multiprotein complexes. Deacetylates SP proteins, SP1 and SP3, and regulates their function. Component of the BRG1-RB1-HDAC1 complex, which negatively regulates the CREST-mediated transcription in resting neurons. Upon calcium stimulation, HDAC1 is released from the complex and CREBBP is recruited, which facilitates transcriptional activation. Deacetylates TSHZ3 and regulates its transcriptional repressor activity. Deacetylates 'Lys-310' in RELA and thereby inhibits the transcriptional activity of NF-kappa-B.
Post-translational
Sumoylated on Lys-444 and Lys-476; which promotes enzymatic activity. Desumoylated by SENP1.
Phosphorylation on Ser-421 and Ser-423 promotes enzymatic activity and interactions with NuRD and SIN3 complexes.
Ubiquitinated by CHFR, leading to its degradation by the proteasome.

Recombinant HDAC1 Antibody (Ab107012) - IF/ICC IF analysis of HDAC1 (green) in HeLa cells. The cells were fixed and permeabilized with 100% methanol for 5 minutes, and blocked with 2% BSA for 1 hour at room temperature. Cells were stained Recombinant HDAC1 Antibody (Ab107012) at 0.25 μg/mL in blocking buffer overnight at 4 °C, and then incubated with Goat Anti-Rabbit IgG H&L (Alexa Fluor® 488) at a dilution of 1/1000 for 1 hour at room temperature in the dark (green). Cells were counterstained with DAPI (blue). Images were taken on the confocal laser scanning microscope.
Recombinant HDAC1 Antibody (Ab107012) - IF/ICC Recombinant HDAC1 Antibody (Ab107012) staining HDAC1 in Hela cells by IF/ICC (immunofluorescence/immunocytochemistry). Cells were fixed with paraformaldehyde, permeabilized with 0.1% Triton X-100 and blocked with 10% goat serum for half an hour at room temperature. Samples were incubated with Recombinant HDAC1 Antibody (Ab107012) (1/500) at 4°C. An Alexa Fluor@ 594-conjugated Goat Anti-Rabbit IgG polyclonal was used as the secondary antibody (1/500). DAPI (blue) was used as the nuclear counter stain. Control: PBS and secondary antibody, An Alexa Fluor® 594-conjugated Goat Anti-Rabbit IgG (1/500).
